CMP-sialic acid synthetase catalyzes the conversion of N-acetylneuraminic acid (Neu5Ac) to CMP-Neu5Ac. The activation of sialic acids (Sia) to CMP-Sia is a prerequisite for the de novo synthesis of sialoglycoconjugates. CSS from Neisseria meningitidis (NmCSS) has been commonly used with other enzymes such as sialic acid aldolase and/or sialyltransferase in synthesizing a diverse array of compounds containing sialic acid or its naturally occurring and non-natural derivatives.

*Enzymatic activity assays are performed in 100 mM Tris-HCl, 10 mM MgCl2, and 500 mM NaCl, pH 8.5 containing Neu5Ac (1 mM) and CTP (1 mM) at room temperature for 5 min and analyzed using HPLC with a UV detector (280 nm).
